Pharmacological Action:

Pharmacodynamics:
Tiamulin is a pleuromutilin-class antibiotic with bactericidal effects at high concentrations. It inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit.

It is particularly effective against:

  • Mycoplasma spp. (chronic respiratory disease in chickens, swine enzootic pneumonia)

  • Brachyspira hyodysenteriae (swine dysentery)

  • Lawsonia intracellularis (proliferative enteropathy)

  • Actinobacillus pleuropneumoniae

  • Staphylococcus and Streptococcus spp. (except group D)

When combined with chlortetracycline at a 1:4 ratio, it is effective in treating mixed infections such as bacterial enteritis and pneumonia in pigs.

Pharmacokinetics:

  • Rapid absorption in pigs with ~85% oral bioavailability

  • Peak plasma concentration in 2–4 hours

  • Highest tissue concentration found in the lungs

  • Metabolized into over 20 active and inactive compounds

  • Excreted mainly via bile in feces, ~30% via urine


Drug Interactions:

  1. Incompatible with ionophores like monensin, salinomycin, and maduramicin – co-use can cause chicken growth retardation, ataxia, paralysis, or death.

  2. Avoid concurrent use with macrolides or lincosamides, as they compete for the same ribosomal binding sites, possibly reducing efficacy.


Indications:

Used for prevention and treatment of:

  • Chronic respiratory disease (CRD) in chickens

  • Swine enzootic pneumonia

  • Swine pleuropneumonia

  • Swine dysentery and proliferative enteropathy caused by Lawsonia intracellularis


Dosage and Administration:

By drinking water:

  • Pigs: 0.45–0.6 g per 1 L, for 5 consecutive days

  • Chickens: 1.25–2.5 g per 1 L, for 3 consecutive days


Adverse Reactions:

Occasional skin redness in pigs. Overdose may cause salivation, vomiting, and central nervous system depression.


Precautions:

  1. Do not use with ionophore antibiotics (e.g., monensin, salinomycin, maduramicin).

  2. Avoid contact with skin and eyes during handling.


Withdrawal Period:

  • Pigs: 7 days

  • Chickens: 5 days
